As both will give a different level with the same car. e.g if i check the oil after 10 mins of engine shutdown at normal operating temp the dip stick reads 1/2 way up, if i then leave the car over night in the same spot and check the oil the following morning the dip stick now reads close to max. which method gives an actual representation of how much oil there is in the sump? This means that any oil thats been distributed around the engine when running has mostly settled back into the sump giving the actual amount.
